Prairie restoration promotes the abundance and diversity of mutualistic arbuscular mycorrhizal fungi.

Predicting how biological communities assemble in restored ecosystems can assist in conservation efforts, but most research has focused on plants, with relatively little attention paid to soil microbial organisms that plants interact with. Arbuscular mycorrhizal (AM) fungi are an ecologically significant functional group of soil microbes that form mutualistic symbioses with plants and could therefore respond positively to plant community restoration. To evaluate the effects of plant community restoration on AM fungi, we compared AM fungal abundance, species richness, and community composition of five annually cultivated, conventionally managed agricultural fields with paired adjacent retired agricultural fields that had undergone prairie restoration 5-9 years prior to sampling. We hypothesized that restoration stimulates AM fungal abundance and species richness, particularly for disturbance-sensitive taxa, and that gains of new taxa would not displace AM fungal species present prior to restoration due to legacy effects. AM fungal abundance was quantified by measuring soil spore density and root colonization. AM fungal species richness and community composition were determined in soils and plant roots using DNA high-throughput sequencing. Soil spore density was 2.3 times higher in restored prairies compared to agricultural fields, but AM fungal root colonization did not differ between land use types. AM fungal species richness was 2.7 and 1.4 times higher in restored prairies versus agricultural fields for soil and roots, respectively. The abundance of Glomeraceae, a disturbance-tolerant family, decreased by 25% from agricultural to restored prairie soils but did not differ in plant roots. The abundance of Claroideoglomeraceae and Diversisporaceae, both disturbance-sensitive families, was 4.6 and 3.2 times higher in restored prairie versus agricultural soils, respectively. Species turnover was higher than expected relative to a null model, indicating that AM fungal species were gained by replacement. Our findings demonstrate that restoration can promote a relatively rapid increase in the abundance and diversity of soil microbial communities that had been degraded by decades of intensive land use, and community compositional change can be predicted by the disturbance tolerance of soil microbial taxonomic and functional groups.

Impact of Silver Nanoparticles on Arbuscular Mycorrhizal Fungi and Glomalin-Related Soil Proteins in the Rhizosphere of Maize Seedlings

Glomalin-related soil protein (GRSP), an important arbuscular mycorrhizal (AM) fungal by-product, plays a key role in preserving or sequestrating soil organic carbon (C). Silver nanoparticles (AgNPs) have become an emerging contaminant and their impacts on soil ecosystems attract increasing concerns. The dynamics of AM fungi and GRSP could therefore form the basis for an in-depth exploration of the influences of AgNPs on soil ecosystems. This study investigated the effects of AgNPs on mycorrhizal growth and AM fungal communities, as well as the GRSP contents in maize (Zea mays L.) soils, with a pot experiment. The contributions of GRSP to soil organic C and the correlations of GRSP with soil organic C were also evaluated. The results indicated that AgNPs decreased the mycorrhizal colonization, AM fungal biomass, and diversity indices, and strongly shifted the community composition of AM fungi with a reduction in Acaulosporaceae and an enrichment in Glomeraceae. Additionally, AgNPs also decreased the soil’s easily extractable (EE) GRSP and total (T) GRSP contents, resulting in lower contributions of EE-GRSP-C and T-GRSP-C to the soil organic C. Linkage analyses revealed that AM fungal abundances have positive correlations with EE- and T-GRSP, and EE- and T-GRSP also positively correlated with soil organic C, indicating that the negative effects of AgNPs on AM fungal abundances and communities were extended to AM-fungal-associated C processes. Altogether, our study found that AgNPs decreased the AM fungal abundances shaped AM fungal communities, and reduced the soil GRSP content, which might subsequently be unfavorable for soil C storage.

Arbuscular mycorrhiza and rhizosphere soil enzymatic activities as modulated by grazing intensity and plant species identity in a semi-arid grassland

Understanding the symbiosis of arbuscular mycorrhizal fungi (AMF) with plants in relation to soil nutrients and enzyme activities under different grazing intensities can be an important guide for the management and protection of semi-arid grasslands. The aim of the present study was to evaluate how the interaction of grazing intensity and plants shapes the composition of AMF communities and enzyme activities in a semi-arid grassland ecosystem in Iran. Sampling focused three dominant plant species (i.e., Salsola laricina, Artemisia siberia, and Stipa hohenackeriana) at sites with different grazing intensities. Soil chemical properties, enzyme activities, root colonization by AMF and AMF communities in the roots were evaluated. Potassium and nitrogen, as well as alkaline phosphatase and urease enzymatic activities were significantly increased at the heavily grazed site, whereas root colonization by AMF was reduced by the high grazing intensity. In addition, AM fungal root colonization is dependent on the host plant species and easier to measure as a sensitive indicator of sustainable grazing. Neither plant species nor grazing intensity affected AM fungal diversity in roots, which could be due to the overall low phylogenetic diversity of AMF in the grassland and the lack of significant differences in soil humidity, pH and organic carbon between the sites. However, plant species and soil properties were the two factors explaining variation in AMF community composition, while grazing had no significant effect. Therefore, AMF communities in root of the semi-arid grassland plants responded largely to plant type rather than to grazing intensity.

Thresholds of Nitrogen and Phosphorus Input Substantially Alter Arbuscular Mycorrhizal Fungal Communities and Wheat Yield in Dryland Farmland.

Arbuscular mycorrhizal (AM) fungi are essential for preserving the multifunctionality of ecosystems. The nitrogen (N)/phosphorus (P) threshold that causes notable variations in the AM fungus community of the soil and plant productivity is still unclear. Herein, a long-term (18 years) field experiment with five N and five P fertilizer levels was conducted to investigate the change patterns of soil AM fungus, multifunctionality, and wheat yield. High-N and -P fertilizer inputs did not considerably increase the wheat yield. In the AM fungal network, a statistically significant positive correlation was observed between ecosystem multifunctionality and the biodiversity of two primary ecological clusters (N: Module #0 and P: Module #3). Furthermore, fertilizer input thresholds for N (92-160 kg ha-1) and P (78-100 kg ha-1) significantly altered the AM fungal community, soil characteristics, and plant productivity. Our study provided a basis for reduced N and P fertilizer application and sustainable agricultural development from the aspect of soil AM fungi.

Arbuscular Mycorrhizal Fungi, Ascophyllum nodosum, Trichoderma harzianum, and Their Combinations Influence the Phyllochron, Phenology, and Fruit Quality of Strawberry Plants

One biostrategy to boost the sustainability of strawberry cultivation is the application of biostimulants to the growing substrate. Here, we investigated whether the use of biostimulants and their combinations affects the strawberry plants’ phyllochron, phenology, and fruit quality. We tested the absence (control) and presence of biostimulants (arbuscular mycorrhizal fungi (AMF), Ascophyllum nodosum (AN), Trichoderma harzianum (TH), AMF + AN, AMF + TH, AN + TH, and AMF + AN + TH). The experimental design used was in completely randomized blocks (four replications). AMF was represented by a multi-species on-farm inoculant; A. nodosum was represented by the commercial product Acadian®; and T. harzianum was represented by the commercial product Trichodermil®. The leaf emission rate, the occurrence and duration of phenological stages, and fruit quality were assessed. The greatest precocity in terms of harvesting the first fruit was observed in plants grown with AMF + TH, which also had the lowest phyllochron (77.52 °C day·leaf−1). Those treated only with AMF were the latest (144.93 °C day·leaf−1). More flavorful fruits were produced by plants grown with AMF + TH. Plants inoculated with the AMF community, whether or not associated with A. nodosum and T. harzianum, had more than 94% mycorrhizal colonization. We conclude that AMF, A. nodosum, T. harzianum, and their combinations influence the phenology, phyllochron, and fruit quality of strawberry plants. In the growing conditions of the Brazilian subtropics, the AMF and T. harzianum combination shortens the strawberry cycle, from transplanting the daughter plants to harvesting the first fruit, and improves the fruit flavor.

Long-Term Application of Biochar Mitigates Negative Plant-Soil Feedback by Shaping Arbuscular Mycorrhizal Fungi and Fungal Pathogens.

Negative plant-soil feedback (PSF) arises when localized accumulations of pathogens reduce the growth of conspecifics, whereas positive PSF can occur due to the emergence of mutualists. Biochar, a carbon-rich material produced by the pyrolysis of organic matter, has been shown to modulate soil microbial communities by altering their abundance, diversity, and activity. For this reason, to assess the long-term impact of biochar on soil microbiome dynamics and subsequent plant performance, we conducted a PSF greenhouse experiment using field soil conditioned over 10 years with Vitis vinifera (L.), without (e.g., C) or with biochar at two rates (e.g., B and BB). Subsequently, the conditioned soil was employed in a response phase involving either the same plant species or different species, i.e., Medicago sativa (L.), Lolium perenne (L.), and Solanum lycopersicum (L.). We utilized next-generation sequencing to assess the abundance and diversity of fungal pathogens and arbuscular mycorrhizal fungi (AMF) within each conditioned soil. Our findings demonstrate that biochar application exerted a stimulatory effect on the growth of both conspecifics and heterospecifics. In addition, our results show that untreated soils had a higher abundance of grape-specialized fungal pathogens, mainly Ilyonectria liriodendra, with a relative abundance of 20.6% compared to 2.1% and 5.1% in B and BB, respectively. Cryptovalsa ampelina also demonstrated higher prevalence in untreated soils, accounting for 4.3% compared to 0.4% in B and 0.1% in BB. Additionally, Phaeoacremonium iranianum was exclusively present in untreated soils, comprising 12.2% of the pathogens' population. Conversely, the application of biochar reduced generalist fungal pathogens. For instance, Plenodomus biglobosus decreased from 10.5% in C to 7.1% in B and 2.3% in BB, while Ilyonectria mors-panacis declined from 5.8% in C to 0.5% in B and 0.2% in BB. Furthermore, biochar application was found to enrich the AMF community. Notably, certain species like Funneliformis geosporum exhibited increased relative abundance in biochar-treated soils, reaching 46.8% in B and 70.3% in BB, compared to 40.5% in untreated soils. Concurrently, other AMF species, namely Rhizophagus irregularis, Rhizophagus diaphanus, and Claroideoglomus drummondii, were exclusively observed in soils where biochar was applied. We propose that the alleviation of negative PSF can be attributed to the positive influence of AMF in the absence of strong inhibition by pathogens. In conclusion, our study underscores the potential of biochar application as a strategic agricultural practice for promoting sustainable soil management over the long term.

Bioprospecting for plant resilience to climate change: mycorrhizal symbionts of European and American beachgrass (Ammophila arenaria and Ammophila breviligulata) from maritime sand dunes.

Climate change and global warming have contributed to increase terrestrial drought, causing negative impacts on agricultural production. Drought stress may be addressed using novel agronomic practices and beneficial soil microorganisms, such as arbuscular mycorrhizal fungi (AMF), able to enhance plant use efficiency of soil resources and water and increase plant antioxidant defence systems. Specific traits functional to plant resilience improvement in dry conditions could have developed in AMF growing in association with xerophytic plants in maritime sand dunes, a drought-stressed and low-fertility environment. The most studied of such plants are European beachgrass (Ammophila arenaria Link), native to Europe and the Mediterranean basin, and American beachgrass (Ammophila breviligulata Fern.), found in North America. Given the critical role of AMF for the survival of these beachgrasses, knowledge of the composition of AMF communities colonizing their roots and rhizospheres and their distribution worldwide is fundamental for the location and isolation of native AMF as potential candidates to be tested for promoting crop growth and resilience under climate change. This review provides quantitative and qualitative data on the occurrence of AMF communities of A. arenaria and A. breviligulata growing in European, Mediterranean basin and North American maritime sand dunes, as detected by morphological studies, trap culture isolation and molecular methods, and reports on their symbiotic performance. Moreover, the review indicates the dominant AMF species associated with the two Ammophila species and the common species to be further studied to assess possible specific traits increasing their host plants resilience toward drought stress under climate change.

Diversity of Arbuscular Mycorrhizal Fungi of the Rhizosphere of Lycium barbarum L. from Four Main Producing Areas in Northwest China and Their Effect on Plant Growth.

Arbuscular mycorrhizal fungi (AMF) can help plants absorb more mineral nutrients after they colonize plant roots, and the mycelia harmonize the soil structure and physical and chemical properties by secreting compounds. AMF species co-evolve with their habitat's geographic conditions and hosts; this gradually causes differences in the AMF species. By using Melzer's reagent to analyze the morphology and using Illumina Miseq sequencing technology to perform the molecular identification of AMF communities among the four typical L. barbarum planting areas (Zhongning, Guyuan, Jinghe, and Dulan) investigated, the variety of L. barbarum roots and rhizosphere AMF communities was greater in the Zhongning area, and every region additionally had endemic species. The successfully amplified AMF was re-applied to the L. barbarum seedlings. We found that the total dry weight and accumulation of potassium increased significantly (p < 0.05), and the root volume and number of root branches were significantly higher in the plants that were inoculated with Paraglomus VTX00375 in the pot experiment, indicating that AMF improves root development and promotes plant growth. We have investigated AMF germplasm species in four regions, and we are committed to the development of native AMF resources. The multiplication and application of AMF will be conducive to realizing the potential role of biology in the maintenance of agroecology.

Impacts of Managed Vegetation Restoration on Arbuscular Mycorrhizal Fungi and Diazotrophs in Karst Ecosystems.

The crucial functional arbuscular mycorrhizal fungi (AMF) and diazotrophs play pivotal roles in nutrient cycling during vegetation restoration. However, the impact of managed vegetation restoration strategies on AMF and diazotroph communities remains unclear. In this study, we investigated the community structure and diversity of AMF and diazotrophs in a karst region undergoing managed vegetation restoration from cropland. Soil samples were collected from soils under three vegetation restoration strategies, plantation forest (PF), forage grass (FG), and a mixture of plantation forest and forage grass (FF), along with a control for cropland rotation (CR). The diversity of both AMF and diazotrophs was impacted by managed vegetation restoration. Specifically, the AMF Shannon index was higher in CR and PF compared to FF. Conversely, diazotroph richness was lower in CR, PF, and FG than in FF. Furthermore, both AMF and diazotroph community compositions differed between CR and FF. The relative abundance of AMF taxa, such as Glomus, was lower in FF compared to the other three land-use types, while Racocetra showed the opposite trend. Among diazotroph taxa, the relative abundance of Anabaena, Nostoc, and Rhizobium was higher in FF than in CR. Soil properties such as total potassium, available potassium, pH, and total nitrogen were identified as the main factors influencing AMF and diazotroph diversity. These findings suggest that AMF and diazotroph communities were more sensitive to FF rather than PF and FG after managed vegetation restoration from cropland, despite similar levels of soil nutrients among PF, FG, and FF. Consequently, the integration of diverse economic tree species and forage grasses in mixed plantations notably altered the diversity and species composition of AMF and diazotrophs, primarily through the promotion of biocrust formation and root establishment.

Common soil history is more important than plant history for arbuscular mycorrhizal community assembly in an experimental grassland diversity gradient

The relationship between biodiversity and ecosystem functioning strengthens with ecosystem age. However, the interplay between the plant diversity - ecosystem functioning relationship and Glomeromycotinian arbuscular mycorrhizal fungi (AMF) community assembly has not yet been scrutinized in this context, despite AMF’s role in plant survival and niche exploration. We study the development of AMF communities by disentangling soil- and plant-driven effects from calendar year effects. Within a long-term grassland biodiversity experiment, the pre-existing plant communities of varying plant diversity were re-established as split plots with combinations of common plant and soil histories: split plots with neither common plant nor soil history, with only soil but no plant history, and with both common plant and soil history. We found that bulk soil AMF communities were primarily shaped by common soil history, and additional common plant history had little effect. Further, the steepness of AMF diversity and plant diversity relationship did not strengthen over time, but AMF community evenness increased with common history. Specialisation of AMF towards plant species was low throughout, giving no indication of AMF communities specialising or diversifying over time. The potential of bulk soil AMF as mediators of variation in plant and microbial biomass over time and hence as drivers of biodiversity and ecosystem relationships was low. Our results suggest that soil processes may be key for the build-up of plant community-specific mycorrhizal communities with likely feedback effects on ecosystem productivity, but the plant-available mycorrhizal pool in bulk soil itself does not explain the strengthening of biodiversity and ecosystem relationships over time.

Plant and soil responses to tillage practices change arbuscular mycorrhizal fungi populations during crop growth.

Tillage practices can substantially affect soil properties depending on crop stage. The interaction between tillage and crop growth on arbuscular mycorrhizal fungi (AMF) communities remains unclear. We investigated the interactions between four tillage treatments (CT: conventional tillage, RT: reduced tillage, NT: no tillage with mulch, and SS: subsoiling with mulch), maintained for 25 years, and two wheat growth stages (elongation stage and grain filling stage) on AMF diversity and community composition. The AMF community composition strongly changed during wheat growth, mainly because of changes in the relative abundance of dominant genera Claroideoglomus, Funneliformi, Rhizophagu, Entrophospora, and Glomus. Co-occurrence network analysis revealed that the grain filling stage had a more complex network than the elongation stage. Redundancy analysis results showed that keystone genera respond mainly to changes in soil organic carbon during elongation stage, whereas the total nitrogen content affected the keystone genera during grain filling. Compared with CT, the treatments with mulch, i.e., NT and SS, significantly changed the AMF community composition. The change of AMF communities under different tillage practices depended on wheat biomass and soil nutrients. NT significantly increased the relative abundances of Glomus and Septoglomus, while RT significantly increased the relative abundance of Claroideoglomus. Our findings indicate that the relative abundance of dominant genera changed during wheat growth stages. Proper tillage practices (e.g., NT and SS) benefit the long-term sustainable development of the Loess Plateau cropping systems.

Characteristics of the Rhizospheric AMF Community and Nutrient Contents of the Dominant Grasses in Four Microhabitats of the Subalpine Zone in Northwestern Yunnan, China

At the southeastern periphery of the Tibetan Plateau, the subalpine ecosystem hosts grasses as some of the most substantial species. However, the community and function of arbuscular mycorrhizal fungi (AMF) around the rhizospheres of grasses in the subalpine zone are still poorly understood. In the present study, 28 soils and 11 species of dominant grasses collected from four microhabitats (shrubland, grassland, woodland, and forest) in the subalpine zone of northwestern Yunnan, China, were used to investigate the AMF community by Illumina MiSeq high-throughput sequencing technology as well as nutrient contents. Among the four microhabitats, the maximum soil nutrient levels around the rhizospheres of grasses were observed in woodland. The nitrogen, phosphorus, and potassium concentrations in Dactylis glomerata shoots were significantly higher than those in the other 10 grass species. The AMF diversity of grassland in summer was substantially greater than that of the other three microhabitats (p &lt; 0.05). Discrepancies were observed within a given plant species across microhabitats; for example, in summer, the nitrogen concentration in the shoot of Iris tectorum in woodland was significantly higher than that in both forest and shrubland (p &lt; 0.05). A total of eight genera were detected in the AMF communities, which were dominated by Glomus, with a relative abundance of 45.4–94.4% in summer and 60.5–84.3% in winter. Moreover, the abundance of Glomus was significantly positively correlated with the content of alkali-hydrolyzable nitrogen in soil and nitrogen in grasses according to the Mantel test. As a critical nutrient element in soil, nitrogen is beneficial for plant growth. Thus, these results provide a better understanding of the resilience of soil AMF community and the ecological adaptability of grasses in the subalpine ecosystems of northwestern Yunnan.

Optimum planting density of Amorpha fruticosa promotes the recovery of understory vegetation and the arbuscular mycorrhizal fungal community in plantations in arid coal mining areas

AbstractEstablishment of plantations may be a useful means of vegetation reconstruction in coal mining areas, especially in fragile environments. However, the influence of shrub planting on the relationship between understory vegetation and soil microorganisms remains poorly understood. Here, we describe a field experiment in which the relationships between understory vegetation and soil microorganisms under different planting densities of Amorpha fruticosa L. (A. fruticosa) were investigated. Compared with areas allowed to recover naturally, shrub planting changed the community and increased the biomass of understory vegetation and soil microbes, especially of arbuscular mycorrhizal fungi (AMF). Planting density was also a key factor that influenced understory vegetation and the soil AMF community and biomass. The maximum diversity of understory vegetation and the AMF community occurred at an intermediate density of introduced shrubs, which was considered to be the optimum density of A. fruticosa. The major soil nutrient cycles that influence understory vegetation and the AMF community were also investigated. An optimum density of introduced shrubs was important in the ecological reconstruction of vegetation in the coal mining area. There may also have been an optimum relationship between the understory vegetation and soil microorganisms especially with AMF in the experimental conditions.

Distinct response patterns of soil micro-eukaryotic communities to early-season and late-season precipitation in a semiarid grassland

Semiarid ecosystems are susceptible to changes in precipitation regimes. However, how soil micro-eukaryote communities in semi-arid ecosystems respond to variations in the seasonal distribution of precipitation remains elusive. A 5-year field experiment was performed to investigate the effect of changed precipitation distributions throughout the growing season on soil micro-eukaryotic communities in the temperate steppe. Early-season precipitation did not significantly affect the biomass of arbuscular mycorrhizal fungi (AMF) nor the composition and structure of AMF and protist communities. Late-season precipitation significantly increased the biomass of AMF while significantly altering the relative abundance of dominant genera in AMF and protist communities. There was a clear difference in the community structure of AMF and protists between the late-season precipitation and natural precipitation. In addition, late-season precipitation decreased the amount of consumer protists but enhanced the abundance of parasitic protists. These results indicate that the impact of precipitation limitations in the middle growing season can be weakened by increased precipitation in the early growing seasons, while an increase in precipitation later in the growing season does not improve the impact of a decrease in precipitation in the middle growing season. Our findings highlight the potential impact of intra-annual changes in precipitation in soil micro-eukaryotic communities in arid and semiarid regions.

Effects of Transformation of Inefficient Camellia oleifera Plantation on Soil Quality and Fungal Communities

Camellia oleifera, a key economic forestry species in southern China, struggles with low productivity due to suboptimal planting management. Recently, transforming old or unadopted varieties of C. oleifera plantations has been recognized as a means to enhance economic benefits and production. However, the impact of these transformations on soil properties and fungal communities has received little attention. In this study, we targeted pre-renewal old C. oleifera and post-renewal young C. oleifera, Pinus massoniana, and Cunninghamia lanceolata. Through field sampling and soil physicochemical property analysis, we developed a soil quality evaluation system that effectively analyzes fungal community structures and identifies key arbuscular mycorrhizal fungi (AMF) species for soil health. We found that the soil quality evaluation system for this region comprises pH, TK, AK, NO3−, PO4− BG, ACP, F.simpson, AMF.shannon, and AMF.ace, which collectively indicated significant improvements in soil quality following transformation. Notably, the nutritional characteristics of the dominant fungal communities underwent marked changes, with an increase in pathogenic fungi in young C. oleifera and an expansion of ectomycorrhizal fungi in P. massoniana forests. The AMF communities in all four types of forest exhibited aggregation, and Scutellospora and Diversispora emerged as key species in the AMF community of C. oleifera. Additionally, Mortierella and Trichoderma were found to enhance plant resistance to pathogenic fungi. This study demonstrates that forestland transformation positively impacts soil quality and fungal community structure in C. oleifera, which provides valuable insights for future soil management in the region, both in terms of soil quality evaluation and fungal conservation.

Year-round dynamics of arbuscular mycorrhizal fungi communities in the roots and surrounding soils of Cryptomeria japonica

Arbuscular mycorrhizal fungi (AMF) live simultaneously inside and outside of host plant roots for a functional mycorrhizal symbiosis. Still, the year-round dynamics and relationships between soil properties and AMF communities of trees in forest ecosystems remain unclear. We collected paired root and soil samples of the same Cryptomeria japonica trees at two forest sites (five trees at each site) every 2 months over a year. Total DNA was extracted from roots and soil separately and soil physicochemical properties were measured. With Illumina’s next-generation amplicon sequencing targeting the small subunit of fungal ribosomal DNA, we clarified seasonal dynamics of soil properties and AMF communities. Soil pH and total phosphorus showed significant seasonality while total carbon, nitrogen, and C/N did not. Only pH was a good predictor of the composition and dynamics of the AMF community. The total AMF community (roots + soil) showed significant seasonality because of variation from May to September. Root and soil AMF communities were steady year-round, however, with similar species richness but contained significantly different AMF assemblages in any sampling month. Despite the weak seasonality in the communities, the top two dominant OTUs showed significant but different shifts between roots and soils across seasons with strong antagonistic relationships. In conclusion, few dominant AMF taxa are dynamically shifting between the roots and soils of C. japonica to respond to seasonal and phenological variations in their microhabitats. AMF inhabiting forest ecosystems may have high environmental plasticity to sustain a functional symbiosis regardless of seasonal variations that occur in the soil.

Dominant herbaceous plants contribute to the spatial heterogeneity of beech and riparian forest soils by influencing fungal and bacterial diversity

Understanding of factors that shape diversity patterns in forest ecosystems is a main challenge in forest ecology and management practices. Although herbaceous plants are known to contribute to the maintenance of the structure and function of temperate forests, their impact on fungal and bacterial communities in soils is largely unknown. Therefore, we conducted a comparative study of soil fungal and bacterial diversity in two Central European deciduous forests, focusing on plots with dominant herbaceous species of contrasting morphology, phenology, reproduction, and ecology, including Allium ursinum and Dentaria enneaphyllos in a beech forest, as well as Aegopodium podagraria and Ficaria verna in a riparian forest. Plots with a mixture of herbaceous species and without plant cover were also studied. In both forests, fungal communities showed the strongest association with spatial location, part of which was explained by between-site variability in soil chemistry and was also influenced by herbaceous vegetation. The community composition of saprotrophic fungi depended on the presence and type of herbaceous vegetation in both forests. In addition, herbaceous plants affected the composition of arbuscular mycorrhizal fungi community in the beech forest, while in the riparian forest they affected endophytes and plant pathogens. For bacteria, soil chemistry played the most important role. Our results showed that dominant herbaceous vegetation is one of the drivers shaping microbial community composition, contributing to spatial heterogeneity of forest sites. Given that herbaceous species, especially those forming monospecific patches, affect the soil biotic properties in temperate forests, they should be included in forest management practices.

Interactions between halotolerant nitrogen-fixing bacteria and arbuscular mycorrhizal fungi under saline stress.

Soil salinity negatively affects crop development. Halotolerant nitrogen-fixing bacteria (HNFB) and arbuscular mycorrhizal fungi (AMF) are essential microorganisms that enhance crop nutrient availability and salt tolerance in saline soils. Studying the impact of HNFB on AMF communities and using HNFB in biofertilizers can help in selecting the optimal HNFB-AMF combinations to improve crop productivity in saline soils. We established three experimental groups comprising apple plants treated with low-nitrogen (0 mg N/kg, N0), normal-nitrogen (200 mg N/kg, N1), and high-nitrogen (300 mg N/kg, N2) fertilizer under salt stress without bacteria (CK, with the addition of 1,500 mL sterile water +2 g sterile diatomite), or with bacteria [BIO, with the addition of 1,500 mL sterile water +2 g mixed bacterial preparation (including Bacillus subtilis HG-15 and Bacillus velezensis JC-K3)]. HNFB inoculation significantly increased microbial biomass and the relative abundance of beta-glucosidase-related genes in the rhizosphere soil under identical nitrogen application levels (p < 0.05). High-nitrogen treatment significantly reduced AMF diversity and the relative abundance of beta-glucosidase, acid phosphatase, and urea-related genes. A two-way analysis of variance showed that combined nitrogen application and HNFB treatment could significantly affect soil physicochemical properties and rhizosphere AMF abundance (p < 0.05). Specifically, HNFB application resulted in a significantly higher relative abundance of Glomus-MO-G17-VTX00114 compared to that in the CK group at equal nitrogen levels. The impact of HNFB on the AMF community in apple rhizospheres is influenced by soil nitrogen levels. The study reveals how varying nitrogen levels mediate the relationship between exogenous HNFB, soil properties, and rhizosphere microbes.
